The Application of MiR-299-5p in the Early Diagnosis and Prognosis Evaluation of Head and Neck Squamous Cell Carcinoma
Objective To evaluate the value of miR-299-5p in the early diagnosis and prognosis of head and neck squamous cell carcinoma (HNSCC).Methods A total of 65 patients with HNSCC who were admitted to our hospital from January,2018 to January,2019 were selected for the study.Cancer tissues and adjacent tissues of these patients were collected during the operation.Fluorescence quantitative PCR was used to detect the level of miR-299-5p in cancer tissues and adjacent tissues.The receiver operating characteristic (ROC) curve was drawn to analyze the early diagnosis of HNSCC by miR-299-5p.Kaplan-Meier method was used to plot the survival curve of HNSCC patients.Cox regression was used to identify the influencing factors of prognosis of patients with HNSCC.Results Compared with adjacent normal tissues,the level of miR-299-5p in cancer tissues of HNSCC patients was significantly reduced (P<0.05).The level of miR-299-5p in cancer tissue of HNSCC patients was correlated with TNM stage,HPV infection,degree of differentiation,and presence or absence of lymph node metastasis (P<0.05),and the AUC of miR-299-5p level for the early diagnosis of HNSCC patients was 0.825,with the sensitivity of 88.51%,and the specificity of 73.83%.The cut-off value was 0.852,and Kaplan-Meier survival curve analysis showed that the 3-year survival rate of miR-299-5p low expression group (9/32,28.13%)was significantly lower than that of miR-299-5p high expression group (23/33,63.89%)(x2=18.698,P=0.003).Univariate Cox regression analysis showed that low expression of miR-299-5p,HPV infection and lymph node metastasis were risk factors for the poor prognosis in patients with HNSCC (P<0.05 ),and multivariate Cox regression analysis showed that low expression of miR-299-5p,HPV infection and lymph node metastasis were independent risk factors for the poor prognosis in patients with HNSCC (P<0.05 ).Conclusion The level of miR-299-5p in cancer tissues of patients with HNSCC is significantly lower than that in adjacent tissues,and it has a high value for the early diagnosis and prognosis of HNSCC.
