The Study of IFN-γ and IL-2 Dual Factor Detection in the Diagnosis of Pediatric Tuberculosis
Objective To explore the diagnostic value of IFN-γ and IL-2 in children with tuberculosis.Methods Between June,2022 and December,2023,89 children with high suspected tuberculosis infection were selected from the Children's Hospital affiliated to Soochow University.The whole blood samples were stimulated with culture filtrate protein 10(CFP10)and early secretory antigenic target-6 kD(ESAT6)for 18-24h,and the supernatant of culture was collected to detect levels of IFN-γ and IL-2 in serum by ELISA method(Define.TB).Results The proportion of children infected with tuberculosis increased with age.In the active tuberculosis group,patients were generally older than 6 years old or younger than 2 years old.In the latent tuberculosis group,patients were generally older than 6 years old;Kappa value of the diagnosis of TB infection by Define.TB and TB-IGRA was 0.785,indicating a good consistency between the two methods;There were significant differences in IFN-γ and IL-2 detection results between active tuberculosis and latent tuberculosis infection groups(P<0.001);ROC curve analysis of the Define.TB showed that the sensitivity,specificity and AUC of the combined diagnosis of IFN-γ and IL-2 were 91.0%,81.8%and 0.880,respectively.Conclusion Combined peripheral whole blood cell IFN-γ and IL-2 dual-factor assay has a high diagnostic value in pediatric tuberculosis,also has good clinical significance for the differential diagnosis of ATB and LTBI.
