A Study of Inhibition and Plasmid Elimination of Scutellaria Scutellaria Against Carbapenemase-producing Klebsiella Pneumoniae
Objective To study the bacteriostasis and plasmid elimination effects of scutellaria baicalensis on carbapenemase-producing Klebsiella pneumoniae(CRKP).Methods The minimum inhibitory concentration(MIC)of scutellaria baicalensis and imipenem on CRKP was determined by broth dilution method.Plasmid elimination rate of CRKP from scutellaria baicalensis was determined by plasmid elimination test.The potential eliminator was identified by automatic microbial mass spectrometry system,while the sensitivity of eliminator to imipenem and meropenem was confirmed by disk diffusion method(K-B method),the production of carbapenase was confirmed by mCIM/eCIM assay,and the blaKPC gene was detected by PCR.Results MIC value of scutellaria baicalensis for CRKP was 6.25mg/mL,and the plasmid elimination rates were 0%,0.3%and 1%at 24,48 and 72 hours,respectively.The eliminator was identified as Klebsiella pneumoniae(KPN)by automatic microbial mass spectrometry system,and they were susceptible to imipenem and meropenem,negative for carbapenem inactivation test(mCIM)/ethylenediamine tetraacetic acid carbapenem inactivation test(eCIM).KPC carbapenase gene(blaKPC gene)was not detected by polymerase chain reaction(PCR).Conclusion Scutellaria baicalensis shows certain level of inhibitory effect and plasmid elimination on CRKP.
