Effect of forsythoside A on expression of intracellular receptors and antiviral gene in IBV-infected cells
[Objective]The aim of this study was to evaluate the anti-infectious bronchitis virus (IBV) activity of forsythoside A and to investigate the mechanism involved in its antiviral action.[Methods]Cell viability was measured by MTT.The IBV-infected HD11 cells were treated with forsythoside A at different concentration (0-20μmol/L) for 36 hours,then Quantitative Real-time PCR was used to determine IBV N gene,intracellular receptors (MDA,LGP2 and NLRC5) and antiviral gene (IRF7,IFN-α and IFN-β) mRNA expression.[Results]Forsythoside A significantly inhibited IBV infection and up-regulated intracellular receptors (MDA,LGP2 and NLRC5) and antiviral gene (IRF7,IFN-α and IFN-β) mRNA levels.[Conclusion] Forsythoside A exerts anti-infectious bronchitis virus activity and activates intracellular receptors (MDA,LGP2 and NLRC5) and antiviral gene (IRF7,IFN-α and IFN-β) in IBV-infected cells.
