Optimization of transient transformation system in peach callus and fruit
[Objective]In order to establish and optimize the transient transformation system of peach callus and fruit.[Methods]The optimal explants and culture conditions for callus induction of peach were screened,and the system of Agrobacterium tran-sient transformation of peach callus and fruit in vitro was optimized.[Results]The leaves,stems and embryos were used as ex-plants to induce callus.The highest induction rate of leaves was 85.8%,and the induction rate of callus induced by embryo was 95.8%,and the formed callus was looser.In subsequent subcultures,PVP treatment with 4 g/L can alleviate the browning of callus.The peach callus was successfully transformed by immersion method,and the fruit block could be successfully trans-formed by vacuum and injection method.The best condition for fluorescence detection was co-culture for 3 days with different transformation methods.[Conclusion]The induction conditions of peach callus were optimized and a relatively stable peach tran-sient transformation system was established,which laid a technical foundation for the rapid verification of peach genes.
