基于高效液相色谱法的苦参指纹图谱研究

[目的]旨在建立苦参高效液相色谱指纹图谱。[方法]采用Waters Atlantisⓒ T3色谱柱(250 mm× 4。6 mm,5。0 μm),流动相为0。01 mol/L乙酸铵(0。045%浓氨水)和乙腈,流速1。0 mL/min,梯度洗脱,色谱柱温度30℃,检测波长225 nm。检测12批苦参样品,建立指纹图谱,进行相似度评价、聚类分析和主成分分析。[结果]建立了苦参的高效液相色谱指纹图谱,苦参样品的相似度在0。940~0。998之间,共标定出23个共有峰,并通过标准品确认了其中5个,分别为氧化苦参碱、氧化槐果碱、槐定碱、苦参碱和槐果碱。聚类分析将12批苦参样品分为4类,主成分分析后23个共有峰缩减为4个主成分。[结论]该方法快速可靠,稳定性和重复性好,可用于苦参的质量控制。
Study on fingerprint of Sophorae flavescentis based on high performance liquid chromatography
[Objective]To establish fingerprints of Sophorae flavescentis Radix by high performance liquid chromatography.[Methods]The chromatographic separation was performed on a Waters Atlantis © T3(250 mm × 4.6 mm,5 μm)analytical column.The mobile phases were 0.01 mol/L ammonium acetate with 0.045%ammonium hydroxide and acetonitrile at a flow rate of 1.0 mL/min with gradient elution.The column temperature was 30 ℃ and the detection wavelength was 225 nm.12 batches of samples were detected,and the fingerprint was established.Similarity evaluation,cluster analysis and principal com-ponent analysis were conducted.[Results]The results showed that high performance liquid chromatography fingerprints for So-phorae flavescentis Radix were successfully established,and similarities of their fingerprints were between 0.940 and 0.998.12 batches of Sophorae flavescentis Radix possessed 23 common peaks and 5 common peaks could be identified using standard products,which were oxymatrine,oxysophocarpine,sophoridine,matrine and sophocarpine,respectively.They were divided into 4 groups by cluster analysis,and 23 common peaks were reduced into 4 principal components after principal component analy-sis.[Conclusion]The method is fast,reliable,stable and reproducible,and can be used for quality control of Sophorae flavescen-tis Radix.
