平谷区马坊镇番茄病毒病的病原鉴定

[目的]为了给平谷区马坊镇设施番茄病毒病防治提供依据。[方法]2022年10月在河北村、果各庄、早立庄、梨羊村、小屯村番茄种植区采集到表现有黄化、蕨叶、褪绿等疑似病毒病症状的番茄叶片,利用小RNA深度测序技术对混合叶片进行病毒筛查,并利用PCR/RT-PCR验证。[结果]侵染当地番茄的病毒种类有黄瓜花叶病毒(cucumber mosaic virus)、南方番茄病毒(southern tomato virus)、番茄褪绿病毒(tomato chlorosis virus,ToCV)和番茄黄化曲叶病毒(tomato yellow leaf curl virus,TYLCV),在 12 份样品中的病毒检出率依次为58。3%、16。7%、16。7%、100%,两种及以上病毒复合侵染率为58。3%,并通过对5个TYLCV进行全序列扩增及构建系统发育树,表明其均与TYLCV北京分离物的亲缘关系最近。[结论]病毒病在马坊镇普遍发生且危害较为严重,检测结果表明,TYLCV是侵染当地番茄的主要病毒,且复合侵染现象严重。
Pathogen(s)identification of tomato viral disease in Mafang Town,Pinggu District
[Objective]To clarify the pathogen of tomato viral disease in Mafang Town,Pinggu District,and provide a basis for the prevention and control of tomato virus disease in this area.[Methods]In October 2022,tomato leaves from Hebei Village,Guogezhuang Village,Zaolizhuang Village,Liyang Village,and Xiaotun Village showing symptoms of viral diseases including yellowing,fern leaves and chlorosis were collected and screened of potential viruses by small RNA deep sequencing,and the i-dentified viruses were further verified by PCR/RT-PCR.[Results]The main virus species infecting tomato plants were cucum-ber mosaic virus(CM V),southern tomato virus(STV),tomato chlorosis virus(ToCV)and tomato yellow leaf curl virus(TYLCV).The detection rates of the corresponding virus in the 12 samples were 58.3%,16.7%,16.7%,and 100%,and the mixed infection rate of two or more viruses was 58.3%,by performing full-genome amplification and constructing phylogenetic trees of 5 TYLCV,it was indicated that all of them are most closely related to the TYLCV isolates from Beijing.[Conclusion]The viral disease was widespread and severe in Mafang town,and the test results indicated that TYLCV was the main virus in-festing local tomatoes,and mixed infection is common.
