Analysis of the role of LsABF4 gene in bolting of leaf lettuce
[Objective]To explore the mechanism of LsABF4 gene in the process of high temperature induced bolting of leaf let-tuce,and to provide theoretical support for the cultivation of anti-bolting leaf lettuce varieties in the future.[Methods]The LsABF4 gene was cloned by homologous cloning using the lettuce variety'GB-30'for easy bolting leaves.Bioinformatics soft-ware and website were used to compare,analyze and predict the encoded amino acid sequence and protein structure of the gene.The role of LsABF4 gene in leaf lettuce bolting was analyzed by virus induced gene silencing technique.[Results]The total length of the CDS sequence of LsABF4 gene was 978 bp,encoding 325 amino acids,the relative molecular weight of the pro-tein was 35 885.21 Da,the theoretical isoelectric point value was 9.47,and the instability index was 48.96.This protein was an unstable protein,and the average protein hydrophobic index was-0.939,which was a hydrophilic protein.The transcrip-tion factor contains the conserved domain of bZIP_plant_BZIP46,and its higher structure is mainly composed of a helix and random coil.After instantaneous silencing of LsABF4 gene,analysis of stem length measurement data showed that stem length of plants in silent group was significantly higher than that in control group and vector group.The results showed that the growth points of the stem tips of the blank control group and the negative control group were conical,and the flower buds were not differentiated,while the stem tips of the experimental group were flat,and the flower buds had entered the differentiation stage.The result indicated that the silencing of LsABF4 by virus induction technique could promote bolting of leaf lettuce.[Conclusion]LsABF4 gene is a typical ABF transcription factor,which has the function of ABF protein family and can inhibit the bolting process of leaf lettuce.
