摘要

[目的]为 β2GPI的获得增加一个新的途径,提高牛血的利用效益,减少牛血资源浪费.[方法]牛血清经HClO4沉淀、Heparin Sepharose CL-6B亲和层析及分子筛等方法纯化获得目的蛋白β2GPI.利用凝胶电泳技术对提取的Bβ2GPI进行了纯度鉴定和分析,然后采用ELISA技术对Bβ2GPI的免疫原性进行了评估.[结果]试验表明提取纯化的Bβ2GPI纯度约为95%,其反应性与β2GPI标准品相同.[结论]从牛血中成功提取纯化β2GPI,且所获得的蛋白具有较高纯度和良好的免疫原性.

Abstract

[Objective]To add a new way to the acquisition of Bβ2GPI,improve the utilization value of bovine blood,and improve the waste of bovine blood resources to a certain extent.[Methods]Initial precipitation with perchloric acid was followed by fur-ther purification using Heparin Sepharose CL-6B affinity chromatography and zeolite.The purity of the extracted Bβ2GPI was identified and analyzed by gel electrophoresis,and then the immunogenicity of Bβ2GPI was evaluated by Enzyme-linked immu-nosorbent assay(ELISA).[Results]Experiments showed that the purity of the extracted and purified Bβ2GPI was about 95%,and its reactivity was the same as that of the β2GPI standard.[Conclusion]Bβ2GPI was successfully extracted and purified from bovine blood,and the obtained protein had high purity and good immunogenicity.
