大黄三味片对便秘大鼠肠道黏膜分泌功能、结构状态及肠道菌群的影响

目的 观察大黄三味片对便秘大鼠肠道黏膜分泌功能、结构状态及肠道菌群的影响.方法 将40只SD大鼠随机分为对照(A)组、模型(B)组、大黄三味片给药7 d(C)组、大黄三味片给药14 d(D)组、大黄三味片给药30 d(E)组,每组8只.B、C、D、E组大鼠先给予盐酸洛哌丁胺8 mg/kg灌胃7 d,造模成功后C、D、E组大鼠分别给予243 mg·(kg·d)-1的大黄三味片混悬液灌胃7、14、30 d.比较A、B、C组大鼠的粪便含水率、肠道分泌功能变化;通过Masson-Fontana黑色素染色法,观察C、D、E组大鼠结肠黑变病发生情况;通过16S rRNA高通量测序分析A、B、C组大鼠肠道菌群的变化;通过液相色谱-串联质谱技术测定A、B、C组大鼠差异代谢物的变化.结果 与A组比较,B组大鼠的粪便含水率、结肠黏膜短路电流下降(P<0.05);与B组比较,C组大鼠的粪便含水率、结肠黏膜短路电流上升(P<0.05).便秘大鼠在经大黄三味片干预7、14、30 d后,结肠黑色素染色实验皆未发现特异性黑色素沉着和嗜银细胞颗粒.与B组比较,C组大鼠颤螺旋菌科(Oscillospiraceae)、普雷沃氏菌科(Prevotellaceae)、毛螺菌科(Lachnospiraceae)和拟杆菌科(Muribaculaceae)的丰度升高.与A组比较,B组大鼠肠道菌群差异代谢物依卡倍特(Ecabet)含量降低(P<0.01);与B组比较,C组大鼠肠道菌群差异代谢物肉桂酸(Cinnamic Acid)含量升高(P<0.05).结论 大黄三味片可通过调节大鼠肠道分泌功能,改善肠道菌群结构和差异代谢物的变化,提高便秘大鼠的粪便含水率,治疗便秘安全且有效.
The effects of Dahuang Sanwei Tablets on secretion function and structure of Intestinal mucosa and gut microbiota in constipated rats
Objective To observe the effects of Dahuang Sanwei Tablets on secretion function and structure of intestinal mucosa and gut microbiota in constipated rats. Methods 40 SD rats were randomly divided into control group (group A),model control group (group B),Dahuang Sanwei Tablets group 7 days group (group C),Dahuang Sanwei Tablets 14 days group (group D) and Dahuang Sanwei Tablets group 30 days group (group E),with 8 rats in each group. Rats in group B,C,D,E were given loperamide hydrochloride 8 mg/kg by Gavage for 7 days. After successful modelling,rats in group C,D,E were treated with Dahuang Sanwei Tablets suspension 243 mg/(kg·d)for 7 days,14 days and 30 days respectively. The curative effect of Dahuang Sanwei Tablets were evaluated by comparing the fecal water content of rats and changes of intestinal secretory function among group A,B,C and the changes of intestinal secretory function were analyzed by Masson-Fontana melanin staining method,the melanosis of colon was observed in rats. 16S rRNA high-throughput sequencing was used to analyze the changes of intestinal flora among group A,B,C and liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used to determine the changes of differential metabolites. Results Compared with group A,the fecal water content and colonic mucosal short-circuit current in group B were decreased significantly(P<0.05);compared with group B,the fecal water content and colonic mucosal short-circuit current of rats in group C were increased significantly(P<0.05).No specific melanin pigmentation and argyrophil granules were found in the colon of constipation rats after 7,14 and 30 days of Dahuang Sanwei Tablets intervention. Compared with group B,the abundance of Oscillospiraceae,Prevotellaceae,Lachnospiraceae and Muribaculaceae was increased in group C.Compared with group A,Ecabet,a metabolite of intestinal flora was significantly decreased in group B (P<0.01). Compared with group B,the content of Cinnamic Acid,the metabolite of intestinal flora,was significantly increased in group C(P<0.05).Conclusion Dahuang Sanwei Tablets can improve the structure of intestinal microflora and the change of differential metabolite by regulating the secretion function of intestinal tract in rats,and increase the water content of feces in rats with constipation with safety and effectiveness.
