龙牙楤木芽多糖提取与脱色工艺的研究

Study on Extraction and Decoloration Technologies of Polysaccharides from Aralia elata (Miq.) Seem.Buds

以龙牙楤木芽为试材,在单因素试验的基础上,以多糖提取率为响应值,通过Box-Behnken试验设计和响应面分析,对龙牙楤木芽多糖提取工艺进行优化;以脱色率和多糖保留率为考察指标,利用正交试验优化活性炭脱除龙牙楤木芽多糖中色素的工艺.结果表明,龙牙楤木芽多糖的最佳提取工艺参数为:提取温度84℃,提取时间5h,液料比26∶1(mL/g),在该工艺条件下龙牙楤木芽多糖平均提取率为5.87%.采用粉末活性炭用量0.6% (m/m)、pH 4.0、温度60℃、时间60 min工艺条件进行脱色处理,平均脱色率达94.08%、多糖保留率为68.15%.
With buds of Aralia elata (Miq.) Seem.as the experimental materials,the extraction conditions of polysaccharide from buds of Aralia elata (Miq.) Seem.were optimized by response surface analysis with the extraction rate of polysaccharide as the response value on the basis of single-factor test and Box-Behnken design of experiment.The decoloration technologies of the polysaccharides with activated carbon were optimized by orthogonal experiment using decoloration rate and polysaccharide retention rate as indexes.The results showed that,the optimum extraction parameters were as follows,extraction temperature was 84 ℃,extraction time was 5 h,liquid to solid ratio was 26∶1(mL/g).Under these conditions,the average extraction rate of polysaccharides from A ralia elta(Miq.) Seem.buds was 5.87%.The optimal decoloration conditions were:activated carbon powder amount was 0.6%(m/m),pH was 4.0,and temperature was 60 ℃,the average decolorization rate was 94.08%,and polysaccharide retention rate was 68.15%.
