Optimization of Ultrasonic-Flash Assisted Extraction Process of Astragalus membranaceus Polysaccharide and Its Antioxidant Activity
Astragalus membranaceus polysaccharide(APS)was extracted from Astragalus membranaceus by ultrasonic-flash assisted extraction.Taking the APS yield as evaluation index,the extraction condition was optimized by single factor test and orthogonal test.The result showed that the optimal ultrasonic extraction conditions were as follows:solid-liquid ratio 1∶30(g/mL),ultrasonic temperature 70 ℃,ultrasonic time 40 min,ultrasonic power 500 W,under these conditions,the APS yield was 7.11%.Based on the optimized result of ultrasonic extraction,combined with flash ex-traction,the optimal flash extraction process paremeters were as folllus:extraction times 3,flash extraction time 100 s,extraction voltage 160 V.And the APS yield was 11.03%.The results of in vitro antioxidant activity experiments showed that when the mass concentration of APS was 1.0 mg/mL,its scavenging rates against 1,1-diphenyl-2-picryl-hydrazyl(DPPH)free radicals and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)cation free radicals were 66.27%and 58.27%,respectively.Which obtained the optimal extraction method for APS with strong antioxidant effects.This method provides a reference for the extraction and development of Astragalus membranaceus polysaccharide.
