Optimization of culture medium and conditions for Morchella mycelium fermentation by response surface
In order to obtain more morchella mycelium and optimize the fermentation medium and culture conditions of morchella mycelium,single factor and response surface optimization method were used to optimize the composition and culture conditions of morchella mycelia liquid fermentation medium.The results show that sucrose is 20 g/L as the best carbon source by single factor test.Yeast extract is selected to be 25 g/L as the best nitrogen source.The optimal inorganic salt is selected as KH2PO4 of 2.0 g/L.The optimal pH value is 7,and the temperature is 28℃.According to the response surface optimization test,mycelium dry weight reaches 17.71 g/L at 25 g/L sucrose,15 g/L yeast paste,2.5 g/L KH2PO4 and 24℃,which is 22%higher than that of mycelium cultured in the original medium.The study lays a foundation for the efficient production of mycelium by morchella liquid and the development of morchella fermented products.
