摘要

采用改良Kennedy's法测定了家蚕精子顶体酶活性,得到了适合家蚕精子顶体酶活性测定的最佳条件,即:孵育温度为25 ℃,孵育时间为4 h;精液体外4 ℃存放随时间延长其顶体酶活性呈下降趋势,酶活性的测定应在15 h内进行.虽然冷冻精液随着冷冻时间的延长其酶活性下降,但是不同冷冻方式之间酶活性变化趋势不同,-80 ℃保存时酶活性直线下降,120 d后酶活性下降了61%.液氮中保存的精液虽然在30 d后的酶活性为冷冻前的58%,但在其后的几个月中酶活性下降不明显.此外,顶体酶活性的高低与精液人工授精产卵数及受精卵率相关,酶活性越低其产卵数越少,受精卵率也越低,提示顶体酶活性的高低可以在一定程度上反映精液的品质.

Abstract

Using the improved Kennedy's method, the acrosin activity of silkworm sperm was determined, and the optium condition for determining the acrosin activity of silkworm sperm was obtained. The incubation temperature is 25 ℃ and incubation time is 4 h. The acrosin activity declined with the time passing when the sperm was preserved at 4 ℃, the determination of acrosin activity should been finished within 15 h. Although the acrosin activity of freezed sperm was dropped with the freezing time passing, the change trends of it were distinct among different freezing modes. When the sperm was preserved at -80 ℃, the acrosin activity was declined in linearity and dropped 61% after 120 d. When the sperm was preserved in liquid nitrogen, the acrosin activity at 30 d is 58 percent of the beginning's, but it dropped unobviously within the following months. Besides, the acrosin activity was related to the total laying eggs number and the fertility rate of artificial insermi-nated sperm. The lower the acrosin activity is, the less the laying eggs number and the lower the fertility rate are. This suggested that the acrosin activity could reflect the quality of sperm to a certain extent.
