Genetic Relationship Analysis of 44 Silkworm Germplasm Resources in Yunnan Province Based on ISSR Molecular Makers
In this study,the genetic relationship of 44 silkworm (Bombyx mori) variety resouces conserved in Yunnan Province was investigated by means of ISSR molecular marking technology to provide the basis for the rational preservation,utilization and selection of parental silkworm varieties of silkworm germplasm resources.Totally 116 bands were amplified from pupal genomic DNA of 44 silkworm varieties using 13 ISSR primers and there were 98 polymorphic bands with a percentage of polymorphic band of 84.5%,implying that there was rich genetic diversity among the tested 44 germplasm resources of silkworm.Genetic similarity coefficients between the 44 silkworm varieties ranged from 0.517 2 to 0.905 2,with an average of 0.711 2.The genetic relationship between these silkworm varieties was analyzed with UPGMA method based on their genetic similarity coefficients.The result showed that the tested silkworm varieties were firstly clustered into Chinese and Japanese strain silkworm varieties,and then further divided into 2 subgroups respectively at genetic similarity coefficient of 0.69 and the silkworm varieties with close genetic relationship were present in the same group.The results of ISSR molecular marking revealed the genetic diversity and genetic relationship of the tested 44 silkworm germplasm resources in Yunnan Province.
