Effects of blackberry lily rhizome preparation on spontaneous activity and Toll-like receptor 7 gene/myeloid differentiation factor 88 signaling pathway in insomnia rats
Objective To explore the effects of blackberry lily rhizome preparation on spontaneous activity,electroencephalogram(EEG),and Toll-like receptor 7 gene(TLR7)and myeloid differentiation factor 88(MyD88)gene expression in brain tissue of PCPA-induced insomnia rats.Methods 70 healthy SD rats were randomly divided into the blank control group,the model control group,blackberry lily rhizome preparation low dose group(26 mg·kg-1),middle dose group(52 mg·kg1),high dose group(104 mg·kg-1),cinnabar anshen pill group(1.62 g·kg-1)and Diazepam tablet group(0.9 mg·kg-1).The relative expression levels of TLR7 and MyD88 genes in each group were detected by Quantitative Real-Time PCR(qRT-PCR).Spontaneous activity of rats was recorded by Etho Vision rat behavioral system.EEG of rats during sleep in each group was recorded by radio telemetry system.Results Compared with the blank control group,the relative expression levels of TLR7 and MyD88 in the brain tissue of the model control group were significantly increased(P<0.01).Compared with the model control group,the mRNA expression levels of TLR7 and MyD88 in blackberry lily rhizome preparation low dose group and middle dose group decreased(P<0.05),and the mRNA expression levels of TLR7 and MyD88 in high dose group significantly decreased(P<0.01).Compared with the blank control group,the exercise amount of rats after PCPA modeling increased significantly;The exercise amount of different doses of blackberry lily rhizome preparation group,cinnabar anshen pill group and Diazepam tablet group was decreased in different degrees,and the exercise amount of blackberry lily rhizome preparation high dose group was decreased significantly.After 120 minutes of administration,compared with the blank control group,the percentage of delta wave in EEG of the model control group decreased(P<0.05);Compared with the model control group,the percentage of delta wave in EEG was increased in blackberry lily rhizome preparation low dose group and middle dose group,cinnabar anshen pill group,and Diazepam tablet group(P<0.05).Conclusion Blackberry lily rhizome preparation can effectively inhibit excitability,increase the percentage of delta wave in EEG,and improve sleep in rats.And this effect may be achieved by regulating the expressions of TLR7 and MyD88 genes in brain tissue.
