Study on the inhibition of microRNA by Wuhu Decoction to improve virus induced asthma airway inflammation
Objective To expolre the immunomodulatory effect of microRNA-144(miRNA-144)on respiratory syncytial virus(RSV)-induced asthma model mice in the process of airway inflammation response and the therapeutic intervention effect of Wuhu Decoction.Methods 40 SPF-grade male C57 mice were randomly divided into the blank group,the model group,Wuhu Decoction group and the Western medicine group,with 10 mice in each group.Except for the blank group,asthma mouse models in other groups were induced by intranasal administration of RSV and intraperitoneal injection of compound chicken ovalbumin(OVA).Wuhu Decoction group was given Wuhu Decoction by gavage,the Western medicine group was given ribavirin + normal saline by gavage,the model group was given equal volume of normal saline by gavage,and the blank group was given equal volume of normal saline by gavage,once a day,for continuous 1 week.After the experiment,the general condition of each group was compared.The DSI Buxco PFT pulmonary function detection system was used to detect the ratio of residual air volume of lung function to total lung volume(RV/TLC),in order to detect the degree of airflow restriction in mice.The pathological changes of the lung tissue in each group were observed by using hematoxylin and eosin(HE)staining,Masson staining and periodic acid-schiff(PAS)staining.The levels of interleukin(IL)-10,IL-17,tumor necrosis factor-α(TNF-α)and transformoing growth factor-β(TGF-β)in bronchoalveolar lavage fluid(BALF)were detected by enzyme linked immunosorbernassay(ELISA)method.;The quantitative polymerase chain reaction(qPCR)method was used to detect the expression of miRNA-144.Results Compared with the blank group,the model group had obvious respiratory symptoms and increased lung function RV/TLC,which all indicated limited airflow,aggravated lung tissue inflammation and damage,changes of inflammatory factors such as IL-10,IL-17,TNF-α and TGF-β in BALF,and upregulated miRNA-144 expression(P<0.05).Compared with the model group,Wuhu Decoction group and the Western medicine group could relieve the respiratory symptoms,lung function ventilation restriction and inflammatory infiltration of lung tissues in asthmatic mice,while expression of miRNA-144 decreased,and the contents of IL-10,IL-17,TNF-α and TGF-β in BALF changed(P<0.05).Conclusion Wuhu Decoction can improve airway inflammation in RSV induced asthma model mice by reducing the expression of miRNA-144 and regulating Th1/Th2 mediated inflammatory response.
