Effect of electroacupuncture on intestinal motility and proinflammatory factor expression in rats with slow transit constipation
Objective To observe the effect of electroacupuncture at"Zusanli"(ST 36)and"Shangjuxu"(ST 37)on intestinal motility and proinflammatory factor expression in rats with slow transit constipation(STC).Methods 24 healthy and clean SD rats were randomly divided into the blank group,the model group,the drug group and the electroacupuncture group,with 6 rats in each group.STC rat models were prepared by gavage of compound diphenoxylate suspension at 10mg·kg-1,once a day,for 14 days.The blank group was given 0.9%sodium chloride solution at the same dose by gavage.The electroacupuncture group received bilateral electroacupuncture treatment for"Zusanli"and"Shangjuxu"points,and selected dense-sparse waves with the frequency of 2/50 Hz,and the current was measured by slight local muscle twitch;Each treatment lasted for 20 min,once a day,for 14 consecutive days.The drug group was treated with suspension of Mosapride citrate tablets by gavage at doses of 1.37 mg·kg-1 and 1 mL·100 g-1,and the treatment course was the same as above.The first black stool excretion time and small intestinal propulsion rate were observed in each group after treatment.The pathological and morphological changes of colon tissue were observed by hematoxylin and eosin(HE)staining,and the expressions of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and CD68 in intestinal tissue were detected by Western-blot.Results The first black stool excretion time in the model group was significantly longer than that in the blank group,and small intestine propulsion rate was significantly decreased(P<0.05).After treatment,compared with the model group,the time of first black stool excretion in the drug group and the electroacupuncture group was significantly reduced,and the small intestine propulsion rate was significantly increased(P<0.05).HE staining showed bleeding in the colonic mucosal tissue of rats after modeling,and damaged histomorphological structure,accompanied by epithelial cell degeneration and necrosis and inflammatory cell infiltration.Electroacupuncture and drug therapy could reduce inflammatory cell infiltration and improve histomorphological changes.Western-blot results showed that the expressions of TNF-α,IL-1β and CD68 in the model group were significantly increased compared with those in the blank group(P<0.05).After treatment,the expressions of TNF-α,IL-1β and CD68 in colon tissue of rats in the drug group and the electroacupuncture group were significantly decreased compared with those in the model group(P<0.05).Conclusion Electroacupuncture can improve the intestinal motional function and repair the pathomorphological changes of colonic tissue in STC rats,and its mechanism may be related to down-regulating the expression of TNF-α,IL-1β and CD68 protein and inhibiting the inflammatory response.
