铷矿物质抑制小鼠肺癌增殖与转移作用及机制研究

目的 研究铷矿物质抑制小鼠肺癌增殖与转移的作用及机制.方法 小鼠皮下注射Lewis肺癌(Lewis lung cancer,LLC)细胞构造肺癌动物模型,药物干预14d后,测量小鼠肿瘤大小、血常规、血清与肿瘤中炎症因子以及活性氧(reactive oxygen species,ROS)表达;观察肿瘤、心、肝、脾、肺、肾等组织病理学改变.小鼠尾静脉注射B16-Fl0细胞构建黑色素瘤肺癌转移模型,检测血常规变化;观察肺湿重及其病理学改变.结果 在铷矿物质对肺癌移植瘤小鼠影响的试验中,药物(铷矿物质或吉非替尼)干预14d后小鼠白细胞(white blood cell,WBC)、淋巴细胞(lymphocyte,Lym)含量均低于模型对照组(P<0.01),且实体瘤体积和重量偏小,肿瘤组织中肿瘤细胞数目减少,细胞核缩小,出现不同程度坏死;心、肝、肺、肾无明显损伤;脾损伤较轻,脾重降低,血清中白细胞介素(interleukin,IL)-6与ROS含量升高,肿瘤坏死因子-α(tumor necrosis factor,TNF-α)含量降低;肿瘤组织中IL-1β、IL-6含量降低,TNF-α含量升高(P<0.05).在铷矿物质对肺癌转移小鼠影响的试验中,药物(铷矿物质或吉非替尼)干预18 d后小鼠血液Neu和WBC水平均显著降低,肺湿重显著降低(P<0.001),肺转移灶少,能明显抑制炎症反应和恶性黑色瘤的肺转移.结论 铷矿物质能够显著抑制小鼠肺癌的增殖和转移,其作用机制可能是通过调节机体免疫,降低炎症因子水平、抑制ROS生成,从而发挥治疗肺癌作用.
Study on the effect and mechanism of rubidium minerals in inhibiting the proliferation and metastasis of lung cancer in mice
Objective To investigate the effect and mechanism of rubidium minerals in inhibiting the proliferation and metastasis of lung cancer in mice.Methods Mice were injected subcutaneously with Lewis lung cancer(LLC)cells to construct the animal model of lung cancer.And after 14 days of drug intervention,the size of tumors,blood routine,inflammatory factors in serum and tumor,and reactive oxygen species(ROS)expression in mice were measured;Histopathological changes in tumor,heart,liver,spleen,lung,kidney were observed.Mouse tail vein injection of B16-F10 cells was used to construct melanoma lung cancer metastasis model,and blood routine changes were detected.The lung wet weight and its pathological changes were observed.Results In the experiment on the effect of rubidium minerals on mice transplanted lung cancer,after 14 days of drug(rubidium minerals or Gefitinib)intervention,the contents of white blood cell(WBC)and lymphocyte(Lym)in mice were lower than those in the model control group(P<0.01);And the volume and weight of the solid tumor were relatively small,the number of tumor cells in tumor tissues decreased,the nucleus shrank,and different degrees of necrosis occured;There was no significant damage to the heart,liver,lungs,or kidneys;Spleen injury was mild,spleen weight decreased,serum levels of interleukin,(IL)-6 and ROS increased,and tumor necrosis factor-α(TNF-α)content decreased;In tumor tissue,the levels of IL-1β and IL-6 decreased,and TNF-α content increased(P<0.05).In the experiment on the effect of rubidium minerals on lung cancer metastasis in mice,after 18 days of drugs(rubidium minerals or Gefitinib)intervention,the contents of neutrophils(Neu)and WBC in mice were significantly reduced,lung wet weight was significantly decreased(P<0.001),there were few lung metastases,and inflammatory response and lung metastasis of malignant melanoma were significantly inhibited.Conclusion Rubidium minerals can significantly inhibit the proliferation and metastasis of lung cancer in mice,and its mechanism of action may play the role in the treatment of lung cancer by regulating the immunity of the body,reducing the levels of inflammatory factors,and inhibiting ROS production.
