Exploration of the effects of Xuanfei Sanjie decoction on tumor inhibition and immune function in lung cancer mice based on intestinal flora--short-chain fatty acids
Objective To explore the mechanism of the effect of Xuanfei Sanjie decoction on tumour inhibition and immune function in lung cancer mice based on intestinal flora--short-chain fatty acids(SCFAs).Methods C57BL/6 male mice were selected to establish a mouse model of lung cancer.Thirty-two successfully modeled mice were selected and randomly divided into the model group(equal volume of 0.9%NaCl solution by gavage),the cisplatin group(intraperitoneal injection of cisplatin solution 0.1 mg·d-1+equal volume of 0.9%NaCl solution by gavage),the Xuanfei Sanjie decoction group(Xuanfei Sanjie decoction by gavage 0.6 mL·d-1)and the combination group(intraperitoneal injection of cisplatin solution 0.1 mg·d-1+Xuanfei Sanjie decoction by gavage 0.6 mL·d-1),with 8 mice in each group.Another eight healthy C57BL/6 male mice were selected as the normal control group.The indexes were tested after 14 days of continuous gavage administration.Results The model group and the cisplatin group showed reduced intake,hair loss and unresponsiveness;the Xuanfei Sanjie decoction group and the combination group had acceptable intake,less hair loss and the more sensitive response.Compared with the model group,the tumour mass was reduced in the cisplatin group,the Xuanfei Sanjie decoction group and the combination group,among which the combination group had the lightest tumour mass and the highest tumour inhibition rate(P<0.05).Compared with the model group,the thymus index,spleen index,Chao1 and shannon decreased in the cisplatin group,and the above indexes increased in the Xuanfei Sanjie decoction group and the combination group,among which the combination group had the higher value(P<0.05).Compared with the model group,the CD4+,CD4+/CD8+and valeric acid and caproic acid contents in the cisplatin group,the Xuanfei Sanjie decoction group and the combination group increased,and the CD8+and acetic acid and propionic acid contents decreased,with the combination group having the highest contents of CD4+,CD4+/CD8+and valeric acid and caproic acid,and the lowest contents of CD8+and acetic acid and propionic acid(P<0.05).Conclusion Xuanfei Sanjie decoction can improve immune function and inhibit tumour growth in lung cancer mice by improving intestinal flora and SCFAs metabolism.
