Study on the characteristics of pain changes in rats within 24 hours after early massage
Objective To evaluate the changes of pain within 24 hours and the optimal time point of efficacy of minor chronic constriction injury(Minor CCI)model rats after the initiation of early massage analgesia through the changes of mechanical and temperature injury perception,so as to provide support for efficacy of the initiation of massage analgesia.Methods 49 SD rats were randomly divided into 7 groups,namely the sham operation group,the model group,the immediate effect group after massage,6 hours group after massage,12 hours group after massage,18 hours group after massage and 24 hours group after massage.Minor CCI model was established in the model group and the massage subgroups.Each subgroup of massage underwent one intervention of three traditional Chinese medicine treatments methods and three points at 7 days after the modeling.Grip intervention was performed in the model group and the sham operation group.After the intervention,cold sensitivity threshold(CST),paw withdrawal mechanical threshold(PWMT)and thermal withdrawal latency(TWL)were tested for different types of pain in each subroup of massage,the model group and the sham operation group.The serum of the patients was taken,and enzyme linked immunosorbent assay(ELISA)was used to test the changes of levels of interleukin(IL)-10 and tumor necrosis factor-a(TNF-α).Model was taken and dorsal root ganglion(DRG)was measured by real-time quantitative polymerase chain reaction(Real-time qPCR).The changes of expression levels of TNF-α,TRPV1 and TRPA1 genes were observed.Results Behavioral tests showed that CST,PMWT and TWL all decreased or prolonged to varying degrees;ELISA detection showed the significant decrease in serum IL-10 and TNF-α levels in rat;qPCR detection showed that the expression levels of TNF-α,TRPV1 and TRPA1 genes decreased.But the results of three detection methods showed significant changes in indicators at different time pints.Conclusion For the three different injury sensitivity thresholds of CST,MWT,and TWL,the significant time points of the analgesic efficacy within 24 hours after massage intervention are different.Massage can exert immediate and sustained analgesic effects by reducing the expression levels of IL-10,TNF-α,TRPV1 and TRPA1.
