Study on the synergistic inhibition of proliferation of non-small cell lung cancer by Shenqi Fuzheng injection combined with cisplatin
Objective To explore the effect of the synergistic inhibition of proliferation of non-small cell lung cancer(NSCLC)by Shenqi Fuzheng(SQFZ)injection combined with cisplatin(DDP).Methods MTT colorimetric method and flow cytometry were used to determined the inhibitory effect and apoptosis of A549/LLC cells.Morphology was assessed by immunofluorescence.Apoptosis related proteins were detected by Western blot.Anti-tumor activity was determined by subcutaneous tumor-bearing mouse model.Results SQFZ injection had no inhibitory effect on tumor cells,but when used in combination with DDP,it showed synergistic or enhanced anti-tumor effects both in vitro and in vivo.Immunofluorescence results showed that combination of SQFZ and DDP could enhance the destructive effect of DDP on tumor cell membrane.In vivo,tumor weights of tumor-bearing mice significantly decreased,and thymus/spleen index and the level of interleukin-2(IL-2)increased after treatment with SQFZ and DDP.Further study indicated that the anticancer effects mentioned above were associated with upregulation expression of Caspase family and Bax/Bcl-2,and inhibition of nuclear factor kappa-B(NF-κB)and vascular endothelial growth factor(VEGF)pathways.Conclusion SQFZ injection can improve the efficacy of DDP and enhance the immunity of the body.
