Cultivars (Grmplasms) Discrimination and Seed Purity Assessment of Grass Elymus sibiricus Using Genomic SSR Markers
Molecular fingerprinting maps of 3 Elymus sibiricus cultivars and 7 germplasms were carried out using 6 simple sequences repeat (SSR) markers developed from the genomic DNA of E.sibiricus.A total of 21 polymorphic SSR fragments were detected in all the tested materials,and 2 to 5 alleles were detected by each pair of SSR primer with an average of 3.5.All the 3 cuhivars and 7 germplasms could be well distinguished by the fingerprinting maps constructed by 3 pairs of core primers.In addition,seed purity assessment was conducted in cultivars ‘Tongde’ and ‘Duoye’ with 3 pairs of SSR primers.It showed that seed purity and the polymorphism of SSR alleles could be effectively identified.Moreover,it revealed that ‘ Tongde’,a domesticated cultivar,had much lower purity and higher polymorphic alleles than ‘ Duoye’,a breeding cultivar.The SSR markers used in this study can be a powerful tool for cultivars or germplasms identification,seeds purity assessment in E.sibiricus.
