Using Microwave Treatment to Construct Endophyte-free Melica transsilvanica Population
In this study mid-low microwave level treatment was used to construct endophyte-free populations of Melica transsilvanica.The elimination effects of the microwave on endophyte were tested by microscope test combined with Endophyte separation,seeds germination index and plant growth index.The result showed that 30 s microwave treatment was effective in killing the endophyte in the seeds.At the same time,there were no obvious adverse effects on the seeds germination rate,germinating potential and germination index.10 s and 20 s treatment in microwave could promote plant growth and decrease endophyte infection rate by 10.7% and 25%,respectively.However,longer time treatment,40 s and more treatment time could inhibit the growth of plant.Therefore,microwave method was an effective way to construct endophyte-free populations of Melica transsilvanica.
