Effect of Electroacupuncture on Insulin Resistance and Inflammatory Factors in Type 2 Diabetes Mellitus Rats
Objective:To observe the effects of electroacupuncture on blood glucose,insulin resistance(IR),and inflammatory fac-tors in type 2 diabetes mellitus(T2DM)rats,and to explore the potential mechanisms of electroacupuncture in treating T2DM.Meth-ods:24 male SD rats were randomly divided into blank group,model group,and electroacupuncture(EA)group,with 8 rats in each group.Except for the blank group,all rats were fed with high-fat and high-sugar diet for 4 weeks and then injected with a small dose of streptozocin(STZ)to establish a type 2 diabetes rat model.The EA intervention selected acupoints of Zusanli,Sanyinjiao,and Xia Wanxiu,using continuous wave,frequency of 10 Hz,intensity of 1.5mA-2 mA,needle retention time of 30 minutes,6 times per week,and a total of 4 weeks of treatment.The body weight and fasting blood glucose(FBG)of each group of rats were observed week-ly.After the intervention,the fasting insulin(FINS),homeostasis model assessment-insulin resistance(HOMA-IR),and morphologi-cal changes of pancreatic tissue in each group of rats were detected.ELISA was used to detect the contents of inflammatory factors such as monocyte chemotactic protein-1(MCP-1),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β),TGF-β1,and interleukin-10(IL-10)in serum of each group.Results:Compared with the blank control group,the model group of rats showed a significant increase in fasting blood glucose(FBG)and Homeostasis Model Assessment of Insulin Resistance(HOMA-IR)(P<0.01).Histological examination(HE staining)revealed irregular pancreatic islet morphology with blurry edges,reduced and disorganized β-cells within the islets,and significantly increased levels of pro-inflammatory cytokines MCP-1,TNF-α,and IL-1β in the serum(P<0.01).In comparison with the model group,the electroacupuncture group of rats exhibited a decrease in FBG and HOMA-IR levels(P<0.05,P<0.01).HE staining of pancreatic tissue in the electroacupuncture group showed clear islet bounda-ries,relatively intact structure,restored islet area and β-cell morphology,and an increased number of β-cells compared to the model group.Furthermore,the expression levels of pro-inflammatory cytokines MCP-1,TNF-α,IL-6,and IL-1β in the serum were effec-tively reduced(P<0.05).Additionally,the levels of anti-inflammatory factors TGF-β1 and IL-10 were significantly elevated(P<0.01,P<0.05).Conclusion:Electroacupuncture can significantly improve the general symptoms of T2DM rats,reduce blood glucose levels,improve insulin resistance,and regulate the expression of inflammatory factors.
