Box-Behnken Response Surface Method Combined with Multi-index Comprehensive Scoring Method was Used to Optimize the Extraction Process of E′seye Formulation Granules
Objective:The optimum extraction conditions of the formula granules of Malus toringoides(Rehd.)Hughes were estab-lished.Methods:The contents of chlorogenic acid,hypericin,isoquercetin,quercetin,rhizoside,quercetin and phloretin in the leav-es of E′seye were determined by HPLC.Based on single factor test,with soaking time,solid-liquid ratio,extraction time and extraction times as factors,using comprehensive scores of the contents of above seven components,total flavonoids,paste yield as indexes.the ex-traction technology of Malus toringoides(Rehd.)Hughes formulation granules was optimized by Box-Behnken response surface method-ology.Results:The optimal extraction technology was to add water to decoction pieces of E′seye for two times,the first time adding 15 times the amount of water,soaking for 30 min,decocting for 45 min;Add 13 times the amount of water for the second time,fry for 45 min,strain,combine the decoction,and get.After 3 times of validation tests,the average comprehensive score was 0.91(RSD =0%,n =3),relative error of which with predicted score(0.938)was 3.08%.Conclusion:Box-Behnken response surface methodology com-bined with multi-index comprehensive score can be used for the extraction technology of Malus toringoides(Rehd.)Hughes formulation granules,and the optimized extraction technology is stable,feasible and reproducible,which could provide a basis for the preparation and industrial production of Malus toringoides(Rehd.)Hughes formula granules.
