目的:探究附子炮制过程中胆巴浸泡工艺对附子生物碱的影响,确定最佳的工艺参数.方法:从胆巴水溶液的浓度、附子在胆巴水溶液中的浸泡时间、附子与胆巴水溶液的料液比三个方面进行单因素考察,参照《中国药典》2020 年版附子的含量测定方法,采用高效液相色谱法对附子中的生物碱含量进行测定,分析附子中生物碱的含量变化.结果:附片中双酯型生物碱含量随着泡胆时间增加、胆巴水溶液料液比减小和胆巴水溶液浓度增大,总体呈下降趋势;单酯型生物碱的含量变化无明显规律;总生物碱含量均呈下降趋势.综合附子的形态和生物碱含量,确定胆巴浸泡的最佳工艺为 35%-40%胆巴水溶液浓度,以 1∶2 的料液比浸泡附子不少于20 d.结论:胆巴浸泡能有效保存附子,并能够降低其有毒成分双酯型生物碱的含量,优选的浸泡工艺可为胆巴炮制附子规范提供参考.
Process of Soaking Fuzi in Solution of Danba
Objective:To explore the effect of Danba soaking process on aconite alkaloids in the processing of Fuzi,and determine the best process parameters.Methods:A single factor investigation was carried out from the three aspects of the concentration of the Dan-ba aqueous solution,the soaking time of the Fuzi in the Danba aqueous solution,and the solid-liquid ratio of the aconite and the Danba aqueous solution.The HPLC method was used to determine the content of the Fuzi according to the "Chinese Pharmacopoeia"2020 e-dition.The alkaloid content in the aconite was determined,and the change of the alkaloid content in the aconite was analyzed.Re-sults:The content of diester alkaloids in Fuzi showed a general downward trend with the prolongation of bile infusion time,the decrease of the solid-liquid ratio of bile solution and the increase of the concentration of bile solution;the content of monoester alkaloids had no obvious change;The total alkaloid content showed a downward trend.Based on the morphology and alkaloid content of Fuzi,the best process for soaking Danba was determined to be 35%-40%Danba aqueous solution concentration,and the aconite soaked at a solid-liquid ratio of 1∶2 for no less than 20 days.Conclusion:Danba soaking can effectively preserve Fuzi and reduce the content of its toxic components,diester-type alkaloids.The optimal soaking process can provide a reference for the processing of Fuzi in Danba.
