氢溴酸樟柳碱对偏头痛大鼠作用的试验研究

目的:研究氢溴酸樟柳碱对硝酸甘油致偏头痛模型大鼠的作用及其作用机制.方法:将60 只大鼠随机分为正常对照组(生理盐水)、模型对照组(生理盐水)、阳性对照组(佐米曲普坦,1.67 mg/kg)、氢溴酸樟柳碱高、中、低剂量组(1.2、0.6、0.3 mg/kg)6 组,除阳性对照组灌胃给药外,其余各组尾静脉注射给药,连续3d;除正常对照组外,其余各组大鼠右肩部皮下注射硝酸甘油注射剂10 mg/kg体重,建立大鼠偏头痛模型,观察行为学指标及通过酶联免疫吸附法(ELISA)、一步法检测大鼠血清及脑组织中 5-羟色胺(5-HT)、去甲肾上腺素(NE)、内皮素(ET-1)、一氧化氮(NO)水平.结果:偏头痛模型大鼠与正常对照组相比较,甩头次数增加;血清及脑组织内5-HT水平下降(P<0.05),氢溴酸樟柳碱中、低剂量组能明显升高偏头痛模型大鼠血清、脑组织内5-HT水平(P<0.05);模型组大鼠血清中ET-1 水平升高(P<0.05),氢溴酸樟柳碱高、中、低剂量组能降低模型大鼠血清中ET-1 水平(P<0.05);模型组大鼠血清及脑组织NE水平,脑组织ET-1 水平与正常对照组比较差异无统计学意义,但氢溴酸樟柳碱可升高模型大鼠血清内NE水平及脑组织内NE、ET-1 水平,个别剂量组与模型组比较差异有统计学意义(P<0.05);与正常对照组相比,模型组大鼠血清NO水平明显升高(P<0.01),与模型组比较,氢溴酸樟柳碱中剂量组可降低模型大鼠NO水平(P<0.05),其余各组有降低其水平的趋势(P>0.05).结论:氢溴酸樟柳碱对硝酸甘油致偏头痛大鼠有治疗作用,其机制可能与调节偏头痛大鼠血清及脑组织内5-HT、NE、ET-1、NO水平有关.
Effect of Anisodine Hydrobromide on Rats with Migraine
Objective:To study the effect of Anisodine Hydrobromide(AH)on the rats with migraine.Methods:Sixty rats were stochasti-cally divided into 6 groups including the model control group,the normal control group,the zolmitriptan control group(1.67 mg/kg),the high-dose(1.2 mg/kg),the medium-dose(0.6 mg/kg)and the low-dose group(0.3 mg/kg).The appropriate drugs were given to the higy-dose,medium-dose and low-dose group by caudal vein injectionwhile the zolmitriptan control group by intragastric administra-tion.After 3 days of administration,the rats were injected subcutaneously nitroglycerin injection 10 mg/kg body mass to establish rat model of migraine in addition to the rats of normal control group.The content of 5-hydroxytryptamine(5-HT),endothelin(ET),norep-inephrine(NE),NO in the serum and brain were determined.Results:Compared with the normal control group,the times of shaking head increased and the level of 5-HT was decreased(P<0.05)in serum and brain of the migraine model rats,the content of ET and NO increased(P<0.05)in the serum.But the content of NE didn't have significantly changes.The AH can improve the active sub-stance content of migraine rat model caused by nitroglycerin(NTG).It can increase the content of 5-HT,NE in serum and that of 5-HT,NE and ET in the brain tissue(P<0.05).Conclusion:AH have a positive effect on the rats with migraine by regulating the con-tents of the brain neurotransmitters.
