Effect of Modified Yinqiao Mabo Powder on Radiation Oral Mucositis in Mice
Objective:To investigate the preventive and therapeutic effect of Modified Yinqiao Mabo powder on radiation-induced oral mucositis(RIOM)mice,and the possible mechanism of the drug.Methods:Forty C57 mice aged 6-8 weeks were randomly divided into blank group,control group,model group and treatment group,with 10 mice in each group.To establish the RIOM mouse model,the head and neck of mice were irradiated with 8 Gy for 3 consecutive days.The control group was treated with non-irradiation and in-tragastric administration of Chinese medicine.The model group was treated with irradiation and intragastric administration of saline.Prophylactic administration was given by gavage from 3 days before radiotherapy until the day of sampling.Body weight was monitored,and the severity of RIOM was evaluated according to toluidine blue staining solution and H&E staining.Cell proliferation and apoptosis were detected by immunohistochemistry and immunofluorescence.Network pharmacology was used to analyze the signaling pathways re-lated to Yinqiao Attenuation particles and detect the expression of related proteins.Results:Compared with the blank group,the total body weight of the model group was decreased(P<0.01).The area of mucositis and ulcer in the total tongue area increased(P<0.01).The thickness of tongue epithelial cells and the number of basal cells were decreased(P<0.01).The expression of tongue proliferation marker protein PCNA was decreased(P<0.01).The expression of apoptotic cells in tongue was increased(P<0.01).The protein expressions of PI3K and AKT related to the disease were increased(P<0.01).Compared with the model group,the a-bove indicators of the treatment group were improved(P<0.01).Conclusion:The preventive treatment of Modified Yinqiao Mabo powder is beneficial to the healing of RIOM,such as reducing ulcer size,increasing epithelial cell thickness and basal layer epithelial cell composition,increasing epithelial cell proliferation and reducing cell apoptosis.Modified Yinqiao Mabo powder may inhibit the ra-dioactive oral mucosal inflammation by down-regulating the expression of PI3K and AKT.
