互作转录组测序技术在感染性疾病研究中的应用现状

摘要
伴随着第二代、第三代测序技术的发展,转录组测序技术的测序深度不断增加,并且在现今的临床医学诊断和治疗中的应用越来越广.互作转录组测序技术(dual host-pathogen ribonucleic acid sequencing,Dual RNA-seq),可以同时提取2个(或多个)物种的RNA进行基因表达的互作分析,得到它们相互作用的机制.在医学上可用于分析感染患者或实验样本与病原体之间转录组水平的互作情况,从而评估患者或实验样本基因表达状态并可用于药物预测.本文主要综述了Dual RNA-seq的工作原理及其在传染病领域的应用,并介绍了其与单细胞测序和非编码RNA分析结合的进展,以期对感染相关的临床和研究工作提供参考.
Abstract
With the development of Next-generation sequencing(NGS) and the Third-generation sequencing(TGS) technology, the sequencing depth of transcriptome sequencing technology is increasing, and it is widely applicated in clinical diagnosis and treatment . Dual RNA-seq can extract the RNA of two (or more) species at the same time for interaction analysis of gene expression, and reflect the mechanism of their interaction, and can be used in medicine to analyze the interaction of the transcriptome between infected patients or experimental samples and pathogens. This allows to assess the gene expression status of patient or experimental samples and use them for drug prediction. This review summarizes the working principle of Dual RNA-seq and its application in medicine, and introduces its progress in combination with single-cell sequencing and non-coding RNA analysis, in order to provide some references for clinical and research work related to infections.
