Clinical significance and diagnostic value of long non-coding RNA LOC152742 expression in peripheral blood of patients with active pulmonary tuberculosis
Objective To investigate the clinical significance and diagnostic value of LOC152742 expression of long non-coding RNA in peripheral blood of patients with active pulmonary tuberculosis(APTB). Methods Collect 70 active pulmonary tuberculosis patients who received treatment in Anhui Chest Hospital from September 2019 to September 2023 as the APTB group,and select pneumonia patients who received treatment in our hospital during the same period as well as health examinations in parallel γ-interferon release tests (IGRAs) showed 50 cases of negative and 50 cases of positive,respectively,serving as the normal control group,pneumonia group,and latent tuberculosis infection group. Collect fasting peripheral blood samples from four groups of study subjects and measure the difference in LncRNA LOC152742 expression in the peripheral blood mononuclear cells (PBMCs). Analyze the relationship between the expression of LncRNA LOC152742 and pathological features in PBMCs of APTB patients. The diagnostic value of LncRNA LOC152742 expression in PBMC for APTB was analyzed using receiver operating characteristic (ROC) curve analysis. Results There was a statistically significant difference in the expression level of LncRNA LOC152742 in PBMCs among the APTB group,latent tuberculosis infection group,pneumonia group,and normal control group (P<0.05). The expression level of LncRNA LOC152742 in PBMCs of APTB group,latent tuberculosis infection group,pneumonia group,and normal control group decreased sequentially,and there was a statistically significant difference between the 2 groups (P<0.05). There was no statistically significant difference in the expression level of LncRNA LOC152742 in PBMCs among APTB patients of different ages,smoking history,and initial/recurrent treatment status (P>0.05). There was a statistically significant difference in the expression level of LncRNA LOC152742 in PBMCs of patients with different concomitant cavities,involvement of lung fields,and concomitant extrapulmonary tuberculosis (P<0.05). The ROC curve shows that the optimal cutoff value for assisting in the diagnosis of APTB by the expression level of LncRNA LOC152742 in PBMC is 1.035,and the AUC is 0.867[95% CI:0.795-0.940],with corresponding sensitivity and specificity of 81.82% and 76.19%,respectively. Conclusion The level of LncRNA LOC152742 in PBMC of APTB patients is abnormally increased,and the level is related to the severity of the disease. Early detection of LncRNA LOC152742 expression level is of auxiliary diagnostic value for APTB.
