Optimization of Extraction Process of Total Flavonoids from Mulberry by Ultrasonic-microwave Assisted Aqueous Two-phase System
Using mulberry fruit as raw material, total flavonoids were extracted by ultrasonic-micro-wave assisted aqueous two-phase system of ethanol and ammonium sulfate.Effects of ethanol mass frac-tion,ammonium sulfate mass fraction, extraction time, microwave power and liquid to solid ratio on the yield were investigated.Based on the single factor experiment, response surface analysis method was used to optimize the experiment and determine the optimal process conditions. Results showed that under the conditions of ultrasonic power(open)of 50 W,ethanol mass fraction of 25%,ammonium sulfate mass fraction of 20%,extraction time of 6.1 min, microwave power of 300 W and liquid to solid ratio of 27∶1(mL/g),the highest extraction yield of flavonoids from mulberry was up to 17.44 mg/g.
