The Expression and Significance of L1CAM in Lung Squamous Cell Carcinoma Based on Bioinformatics Analysis
Objective:To analyze the expression and significance of cell adhesion molecule Ll(L1CAM)in lung squamous cell carcinoma(LUSC)and its potiental bilogical significance.Methods:The UALCAN database was used to analyze the expres-sion of L1CAM in tumor and normal tissues,and real time fluorescence quantitaive PCR(qPCR)was performed to verity the ex-pression of L1CAM innormal cell and tumor cell lines.The relationship between L1CAM expression and the prognosis of LUSC pa-tients was analyzed using the Sangerbox3.0.The co-expressed genes were searched on LinkedOmics.GO and KEGG pathway a-nalysis were used to determine the biological processes and functions of these genes.Finally,the STRING database was used to analyze the interaction network of proteins encoded by related genes.Results:Compared with normal lung tissues,LI CAM ex-pression levels were significantly increased in LUSC tissues patients.As verified by qPCR,compared with normal lung epithelial cells,the expression of L1CAM in lung squamous cell lines was significantly increased.The main biological processes of co-ex-pressed genes include cell development,cellular or subcellular component movement,intracellular signal transduction regulation,etc.The main enriched cell components included extracellular matrix,endoplasmic reticulum cavity,mitochondrial membrane,etc.The main biological functions were involved in signal receptor binding,calcium ion binding,extracellular matrix structural components,etc.The enriched KEGG signaling pathway is mainly related to focal adhesion,ECM receptor interaction,platinum resistance,and other pathways.The OS,DSS,DFI,and PFI of patients with high L1CAM expression were significantly lower than those with low L1CAM expression.PPI network showed that L1CAM protein interacted with EGFR,NCAM1,ANK,and so on.Conclusion:L1CAM is highly expressed in LUSC tissue and can be used as a prognostic marker.Moreover,the expression of the L1CAM gene is involved in many biological functions,so it is expected to be an imporcant biomarker of lung squamous cell carcinoma.
