Nontargeted lipidomics Analysis of Exosomal Lipid Metabolism in Mice with Allergic Asthma
Objective:To investigate the differences of lipid contents of exosome in Bronchoalveolar lavage fluid between mice with allergic asthma,and to reveal the changs of lipidomics in the process of allergic asthma.Methods:Sixty C57BL/6J mice were randomly divided into an HDM group and a control group.The HDM group received HDM nasal drops,while the control group re-ceived the same dose of physiological saline.Two weeks later,BALF and lung tissue were obtained.HE staining and PAS staining were used to observe the morphology of mouse lung tissue.Exosomes were extracted by ultra-high speed centrifugation,identified by scanning ctansmission electron microscopy and nanoparticle tracer analysis,and analyzed by non-targeted lipidomics by mass spec-trometer.Results:AA mice induced by HDM were successfully established.HE staining showed infiltration of inflammatory cells in the lung tissue of HDM group mice,while PAS showed goblet cell proliferation and increased mucus secretion.Giemsa staining showed an increase in inflammatory cells,especially an increase in the percentage of eosinophils and neutrophils.Untargeted lipido-mics were used between the HDM group and the control group.Based on FC values,p-values,and variable importance for the pro-jection(VIP)value obtained from Orthogonal Partial Least Squares Discriminant Analysis(OPLS-DA),a total of 28 differential lip-id compounds were detected.Most of them were glycerides and sphingolipids.The random forest further narrowed down the lipid range and screened the top 5 lipids,namely PS,SPH,Cer,Cer,and PG.These lipids were higher in the HDM group than in the control group,and the difference was statistically significant(P<0.05).Conclusion:There is a difference in the exosomal lipids of bronchoalveolar lavage fluid between allergic asthma mice induced by HDM and normal mice,PS、SPH、Cer and PG may be new tar-gets for allergic asthma prediction and intervention.
