以铁皮石斛(Dendrobium officinale)类圆球茎为研究材料,系统地研究了影响其液体悬浮培养增殖的诸多因素,如不同质量浓度的MS基本培养基、NAA、蔗糖、椰乳以及培养基pH值和接种量,旨在构建铁皮石斛类圆球茎液体悬浮培养增殖体系.研究结果表明:在液体悬浮培养条件下,最有利于铁皮石斛类原球茎的生长增殖的培养基为1/2MS基本培养基中添加1.0 mg/L NAA,50 g/L椰乳,蔗糖30 g/L;接种量为40 g/L.
Establishment of Suspension Culture System of Protocorm-like Bodies for Dendrobium candidum
An experiment was conducted to study the effects of different concentrations of MS culture medium, NAA, sucrose, coconut milk, and inoculation density on proliferation of protocorm-like bodys (PLBs) of Dendrobium officinale in suspension culture. Results showed that 1/2MS medium supplemented with 1.0mg/L NAA, 50g/L coconut milk, and 3 g/L sucrose was optimal for the proliferation of the PLBs. Inoculation with 40 g PLBs in 1 liter of liquid medium was most suitable for PLBs proliferation.
