蓝花楹茎腐病--病原与系统发育1)

在四川省西昌市蓝花楹( Jacaranda acutiof lia)栽培区内,通过固定样方定点定期观察,发现1种侵染蓝花楹茎干基部的真菌性新病害———蓝花楹茎腐病,对其症状进行了记载描述。通过病株采样、分离、单孢培养及致病性测定,获得代表性菌株A1、B1,并对其进行了形态学及基于rDNA-ITS序列的系统发育分析。结果表明:A1、B1菌株的形态学特性分别与镰刀菌属的尖孢镰刀菌( Fusarium xo ysporum)和厚垣镰刀菌( Fusarium chlamydos-porum)一致;利用真菌通用引物ITS1和ITS4进行rDNA的扩增,对扩增产物进行回收、克隆、测序,分别得到2条大小为516、523 bp的基因片段,通过 Blast比对,建立同源性较高的Fusarium属菌株的系统发育树,最终鉴定A1和B1分别为F.oxyspro um和F.chlamydosporum。
Pathogen Identification of Jacaranda acu tifolia Stem Rot and Its Phylogente ic Analysis
By observing the fixed sample plots , we discovered a new fungal disease named Jca arandaa cutifolia stem rot, mainly damaging to the stem of Jacaranda acutifolia Humb .Et Bonpl , on J.acutfi olia planting regions in Xichang , Sichuan Prov-ince, and recorded the symptoms .We collected the junctions of diseased and healthy tissues , and determined isolated , cultured with single spore , and the pathogenicity .We obtained A1 and B1, as two separate dominant strains , by morpho-logical and developmental systems rDNA-ITS sequences .Two strains in morphological characteristics were consistent with Fusarium oxysporum and Fusarium chlam ydosporum,respectively .Using the fungal universal primers ITS 1 and ITS4, the internal transcribed spacer rDNA sequences of them were amplified , then after recyclying , cloning and sequencing the am-plification products , two longth of 516 and 523bp gene fragments were got severally .By the results of Blast in Genbank , the analysis of fungal rDNA systemic development and phylogenetic tree , we confirmed the pathogens of J.acutifoliastem rot and these two fungal pathogen strains A 1 and B1 were identified as F.oxysporum and F.chlamydosporum, respectively.
