楸树与滇楸组培快繁技术

以楸树( QS2)和滇楸( DQ72)的优良单株为试材,开展组织培养研究,以期建立两者高效的组培快繁技术体系,为楸树和滇楸的种苗生产提供参考。结果显示:0.1%氯化汞处理6.5 min适合QS2和DQ72的茎段消毒。接种20 d后,QS2的褐化率和污染率分别为16.67%和19.79%,DQ72分别为18.75%和21.88%。 DKW( Driver and Kuniyuki Walnut )培养基适合作为QS2和DQ72的基本培养基。 QS2和DQ72适宜的初代培养基均为DKW+0.8%琼脂+1 g· L-1活性炭+2.5%蔗糖+1.0 mg· L-16-BA+0.1 mg· L-1 IBA,接种30 d时的腋芽萌发率分别为84.38%和87.50%。 QS2和DQ72适宜的增殖培养基为DKW+0.6%琼脂+2.5%蔗糖+3.0 mg· L-16-BA+0.2 mg· L-1 IBA,接种35 d时的增殖系数分别达到6.88和6.63,且增殖幼苗生长健壮。 QS2和DQ72适宜的生根培养基为DKW+0.8%琼脂+2.5%蔗糖+0.2 mg· L-1 NAA+0.1 mg· L-1 IBA,在该培养基上接种30 d时,QS2和DQ72的生根率均达到100%,QS2的根系数量和根系平均长度分别达到12.1条和5.3 cm,DQ72分别达到18.5条和3.2 cm。 QS2和DQ72的组培苗移栽60 d后的成活率分别为92.3%和96.6%。
Rapid Propagation System of Catalpa bungei and C.fargesii Bur.f.duclouxii
To promoting seedling production , we selected superior individuals of C.bungei ( QS2) and C.fargesii Bur.f.du-clouxii ( DQ72) to establish its effective rapid propagation system .Proper sterilization method of stem segments was treated by 0.1%HgCl2 for 6.5 min.16.67%and 19.79%of browning rate, as well as 18.75%and 21.88%of contamination rate were observed in QS 2 and DQ72 after inoculated for 20 d, respectively .DKW ( Driver and Kuniyuki Walnut ) basic medi-um was suitable for tissue culture of QS 2 and DQ72.The optimum initial culture medium was DKW+0.8%agar+1 g· L-1 activated carbon+2.5%sucrose+1.0 mg· L-1 6-BA+0.1 mg· L-1 IBA indicated by 84.38%and 87.50%of germination rate of axillary buds, respectively, in QS2 and DQ72 after growing for 30 d.The best medium for cluster buds induction was DKW+0.6%agar+2.5%sucrose+6-BA 3.0 mg· L-1+0.2 mg· L-1 IBA.Q2S and DQ72 proliferated strong shoots and displayed multiplication factor of 6.88 and 6.63 after 35-day growth, respectively.The best rooting medium was DKW +0.8%agar+2.5%sucrose+0.2 mg· L-1 NAA+0 .1 mg· L-1 IBA with 100%of rooting rate in both of QS 2 and DQ72 after growing for 30 d.In the medium, QS2 displayed root number of12 .1and root length of 5.3 cm in average.While, in DQ72 these were 18.5 and 3.2 cm, respectively.The survival rate of QS2 and DQ72 were 92.3%and 96.6%, respectively, after trans-planted for 60 d.
