摘要

氨基酸因含有氨基、羧基等活性基团,易于对其结构进行调控,是构筑小分子有机凝胶的常用骨架或切块.以天冬酰胺为基本骨架,将羧酸位点与正己胺进行缩合反应,形成酰胺键,是一种良好的凝胶因子中间体.研究了Boc-L-天冬酰胺与正己胺的缩合反应,考查了有机碱、反应溶剂和反应温度对反应的影响.条件优化之后发现使用DCC作为缩合剂,DIPEA提供碱性环境,25℃下在DCM溶剂中反应,反应收率可达74%.合成的目标化合物结构经过 1H NMR表征确认,该工艺条件温和,操作简单,效率高.

Abstract

Amino acids are commonly used as backbones or cut blocks for constructing low molecule weight organogels because they contain reactive groups such as amino and carboxyl groups which are easy to modulate their structures.With asparagine as the basic backbone,the carboxylic acid site can be condensed with n-hexylamine to form an amide bond,which is a good intermediate for gel factors.The condensation reaction of Boc-L-asparagine with hexylamine was studied.The effects of organic base,reaction solvent and reaction temperature on the reaction were investigated.After optimizing the conditions,it was found that when DCC was used as condensation agent,DIPEA provided alkaline environment,and the reaction was carried out in DCM solvent at 25℃,the reaction yield could reach 74%.The structure of the synthesized target compound was confirmed by 1H NMR.The process has mild conditions,simple operation and high efficiency.
