Research on Quality Control Method of Anthocyanin in Blueberry Based on Fingerprint and Quantitative Analysis of Multi-components by Single Marker
A multi-component content determination method of blueberry anthocyanin extract was established by combining HPLC fingerprint with QAMS.HPLC was used to construct the blueberry anthocyanin extract fingerprint.With cyanidin as an internal reference,the relative correction factors of delphinidin,petunia chloride,peonidin,malvidin and cyanidin were established.The anthocyanin content in ten batches of blueberry extract was determined by external standard method and QAMS respectively to verify the applicability and accuracy of this method.The quality control method for fingerprint of anthocyanin extract of blueberry was established,and the adulterated products were identified.QAMS was established to determine,and the fr/i of five anthocyanins were determined to be 1.09,0.45,0.49 and 0.51,respectively,and had good durability under different chromatographic conditions.The contents of ten batches of extraction were determined,and there was no significant difference between the calculated values of QAMS and external standard method.The mass fractions of anthocyanins in blueberry were 56.44~225.25 μg·mg-1,20.22~61.70 μg·mg-1,20.63~60.36 μg·mg-1,3.74~6.09 μg·mg-1,65.78~123.50 μg·mg-1.The total anthocyanin content of 10 batches of samples was divided into three levels,which was consistent with the results of cluster analysis.The established fingerprint can be used to identify the authenticity of blueberry anthocyanins,and at the same time,five anthocyanins can be quantitatively detected by QAMS.
