凡纳滨对虾肠道中地衣芽孢杆菌的分离鉴定及其生物学特性

Isolation,identification and biological characteristics of Bacillus licheniformis from the intestine of Pacific white leg shrimp(Litopenaeus vannamei)

为筛选对凡纳滨对虾(Litopenaeus vannamei)具有潜在益生效果的菌株,从体质量为(3.1±0.4)g的健康凡纳滨对虾肠道中筛选出一株具有耐高温(80℃)能力的菌株(命名为RHVJ),采用形态学观察、生理生化鉴定、基因序列比对、病原菌拮抗能力测定、安全性与耐受性分析和生长特性检测等方法对其展开研究.结果表明:菌株RHVJ为革兰氏阳性杆菌,综合其生理生化特征可初步鉴定该菌属于芽孢杆菌属(Bacillus);菌株 RHVJ 的 16S rDNA 序列与地衣芽孢杆菌(Bacillus licheniformis)的 MH305331.1 和NR074923.1 完全匹配,其gyrB序列与CP038186.1和CP023729.1 也达到了 100%的一致性;综合菌体形态、生理生化及分子生物学特征,判定菌株RHVJ为地衣芽孢杆菌;地衣芽孢杆菌RHVJ可抑制副溶血弧菌(Vibrio parahaemolyticus)和维氏气单胞菌(Aeromonas veronii)生长,对凡纳滨对虾无致病性;在胆盐质量浓度为 0~1.2 g/L、pH为 3~11 环境下培养,以及在 37~90℃下进行 5 min或 15 min水浴处理后,菌株RHVJ仍可持续增殖,该菌在 27℃、pH 7 和盐度 20 条件下生长最佳.研究表明,凡纳滨对虾肠道源地衣芽孢杆菌RHVJ环境适应性良好且安全,具有一定的病原菌拮抗能力,可作为候选菌株开发为益生菌制剂.
In order to screen strains with potential prebiotic effects on Pacific white leg shrimp(Litopenaeus van-namei),a strain(RHVJ)with high temperature(80℃)tolerance was isolated from the intestine of healthy Pacif-ic white leg shrimp with body weight of(3.1±0.4)g,and identified by morphological observations,physiological and biochemical determination,and gene sequence alignment.The strain RHVJ was detected for pathogen antago-nism,safety,tolerance analysis,and growth characteristics.The results showed that RHVJ was a gram-positive Ba-cillus,and tentatively identified as a Bacillus based on physiological and biochemical characteristics.The 16S rDNA sequence of RHVJ was found to be completely matched with Bacillus licheniformis strains MH305331.1 and NR074923.1,and its gyrB sequence also showed 100%consistency with CP038186.1 and CP023729.1.The strain RHVJ was identified as Bacillus licheniformis,inhibited the growth of Vibrio parahaemolyticus and Aeromonas veronii,and not pathogenic to Pacific white leg shrimp based on physiological and biochemical identification re-sults.When cultured in environments with bile salt concentrations ranging from 0 to 1.2 g/L and pH levels between 3 and 11,and subjected to water bath treatments at temperatures from 37 to 90℃for either 5 or 15 minutes,the RHVJ strain demonstrated sustained proliferation.It exhibited optimal growth under conditions of 27℃,pH 7,and 20 salinity.The B.licheniformis RHVJ from the intestine of Pacific white leg shrimp had good environmental adapt-ability and safety,as well as some ability to antagonize pathogenic bacteria,and were used as a candidate strain for the development of probiotics.The findings laid the foundation for its application in prawn culture.
