不同离子浓度和温度对北方马口鱼精子活力的影响

摘要
为了解中国北方马口鱼(0psariichthys bidens)精子的生理特征并提高其人工繁殖受精率,以精子快速运动时间(fast movement time,FT)、寿命(life span,LT)和激活率(activation rate,AR)作为精子活力的评价指标,通过显微观察法,检测了用不同浓度的Na+、K+、Ca2+、葡萄糖、Tris、甘油精子激活液及水温等处理对马口鱼精子活力的影响.结果表明:单因子精子激活液中,马口鱼在浓度为34 mmol/L Na+、40 mmol/L K+、17 mmol/L葡萄糖、温度为20 ℃条件下,精子快速运动时间、寿命均最高;当Tris缓冲剂浓度为33 mmol/L时,精子快速运动时间达到最大值,当Tris缓冲剂浓度为0 mmol/L时,其精子寿命最长,而在Ca2+和甘油溶液中,精子快速运动时间和寿命均小于对照组(0 mmol/L),其对马口鱼精子活力具有抑制作用;在复合因子精子激活液中,马口鱼精子在激活液B(34 mmol/L Na++40 mmol/L K+,二者体积比为1∶1)中的快速运动时间、寿命均最高,精子活力最强.研究表明,适宜浓度的Na+、K+、葡萄糖和Tris因子对马口鱼精子活力有增强效果,马口鱼精子存活的适宜温度为17～25 ℃;在进行北方马口鱼人工授精时,可使用激活液B(34 mmol/L Na++40 mmol/L K+,二者体积比为1∶1)来提高其精子活力.
Abstract
In order to understand the physiological characteristics of sperm of Chinese hook snout carp(Opsariich-thys bidens)and improve its artificial reproduction fertilization rate,the motility including fast movement time,life span and activation rate was determined in the sperm exposed to different concentrations of Na+,K+,Ca2+,glu-cose,Tris,glycerol and different water temperatures in Chinese hook snout carp.The results showed that the maxi-mal fast sperm motility time and life span were observed in the sperm of Chinese hook snout carp under the condi-tions of Na+34 mmol/L,K+40 mmol/L,glucose 17 mmol/L,and 20 ℃,with the maximal fast motility time at Tris buffer of 33 mmol/L,and the maximal life span at Tris buffer 0 mmol/L.There were shorter fast sperm motility time and life span in Ca2+and glycerol solutions than those in the control group,both showing inhibitory effects on sperm motility in Chinese hook snout carp.The maximal fast movement time and life span were found in the sperm exposed to complex factor sperm activation solution B(34 mmol/L Na++40 mmol/L K+,with volume ratio of 1∶1).In conclusion,suitable Na+,K+,glucose and Tris led to an enhanced effect on sperm motility in Chinese hook snout carp,with suitable temperature of 17-25 ℃.In the artificial insemination of Chinese hook snout carp,activation solution B(34 mmol/L Na++40 mmol/L K+,with volume ratio of 1∶1)is recommended to be used to improve the sperm motility.
