摘要

为探究江西水产养殖系统中细菌资源分布规律及水环境因素对细菌群落的影响,应用 Illumina MiSeq高通量测序技术对江西省鄱阳湖周边多种鱼类养殖池塘水体进行 16S rRNA测序,并探讨了细菌群落多样性及结构特征.结果表明:放线菌门(Actinobacteriota)为优势菌门(19.80%~68.86%),其次主要类群有变形菌门(Proteobacteria)、蓝细菌门(Cyanobacteria)、拟杆菌门(Bacteroidota)、HgcI_Clade、CL500-29_marine_group及Cyanobium_PCC-6307 等;各养殖池塘Shannon指数为 2.47~4.25,平均值为3.31,Simpson指数为 0.03~0.23,平均值为 0.06,Chao1 指数为 140.07~573.17,平均值为 353.23,Ace指数为 146.52~564.34,平均值为 350.73;各站点间细菌群落丰富度、均匀度存在较大跨度;在空间上,细菌群落多样性不存在显著性差异(P>0.05),而对于不同养殖模式,Ace指数与Chao1 指数均表现为混养池塘>单养池塘(P<0.01);相关性分析显示,养殖水体的TSS、DO与细菌α多样性相关性最强,水温(WT)、总氮(TN)、氨氮(NH4+-N)、总磷(TP)、高锰酸盐(CODMn)等环境因素显著影响水体微生物群落结构(P<0.05).研究表明,水环境因素具有改变菌群结构的潜能,可通过适当增氧、提高循环率、增加养殖鱼类种类等措施提高养殖水体细菌多样性,以实现高效养殖.

Abstract

In order to understand the distribution of bacterial resources and influence of aquatic environmental fac-tors on bacterial communities in Jiangxi aquaculture system,the structure and diversity of bacterial communities were explored in central region of various fish aquaculture ponds around Poyang Lake in Jiangxi Province(115°20′21″-117°57′32″E,27°16′38″-28°48′42″N)in June 2022 by sequence 16S rRNA and Illumina MiSeq high-throughput sequencing technology.The results showed that,Actinobacteriota was the dominant bacteria(19.80%-68.86%),followed by Proteobacteria,Cyanobacteria,Bacteroidota,HgcI_Clade,CL500-29_marine_group and Cyanobium_PCC-6307.Shannon index was found to be fluctuated between 2.47 and 4.25,with average value of 3.31.Simpson index was fluctuated between 0.03 and 0.23,with average value of 0.06.The fluctuation of Chao1 index was shown to be changed from 140.07 to 573.17,with average of 353.23.The Ace index in each aquaculture pond fluctuated from 146.52 to 564.34,with average value of 350.73.There were large range of abundance and u-niformity of bacterial communities at each site.Very higher Ace index and Chao1 index were observed in polyculture ponds than those in monoculture ponds in different farming modes(P<0.01),though without significant difference in bacterial community diversity in space.The correlation analysis showed that there was closest relationship be-tween TSS and DO contents and the α-diversity of bacteria,with significant effects of environmental factors such as WT,TN,NH+4-N,TP and CODMn on bacterial community structure in water(P<0.05).It was found that water en-vironment factors had the potential to change the structure of bacterial community,and that the bacterial diversity in cultured water was improved by appropriate measures including increase in oxygen,circulation rate and culture spe-cies,so as to achieve efficient culture.
