Protective effect of Shen Ning Capsules on kidney of type 2 diabetic rats
AIM: To study the protective effect of Shen Ning Capsules on rat kidney of type 2 diabetes mellitus and its mechanism. METHODS: SD rats were randomized into the control, model and treatment groups. Rat models of type 2 diabetes mellitus were established by high carbohydrate and fat food and small dose of streptozotocin (STZ) injection. Shen Ning Capsules were given to the treatment group and purified water to the control and the model groups. After 6 months, serum and urinary biochemical and kidney morphological examination were conducted. The levels of Ang- Ⅱ and endothelin ( ET) in serum were determined by ra-dioimmunoassay and the transforming growth factorβ1 (TGF-β1) in renalcortex was detected by immunohistochemistry. RESULTS : Compared with those in the model group, the blood sugar, 24 h urine protein, BUN and serum creatinine obviously improved in the treatment group (P < 0. 05 ). In the treatment group, the renal hypertrophy index (the right kidney weight /body weight) descended, the renal glomerulus diameter was shortened, mesenterium hyperplasia and the basilar membrane thickening were alleviated and kidney tubules cavity and stromal cells infiltration gradually decreased (P < 0.05). Ang- Ⅱ and ET levels in serum and renal cortex reduced (P < 0.01, P < 0. 05) and the expression of TGF-β1 decreased (P < 0. 05). CONCLUSION: Shen Ning Capsules exert some protective effect on kidney of diabetic rats, which may be related to the inhibition of the over ex- pression of ET and TGF-β1.
