Effect of adenovinls-mediated IL-24 gene on expression of p38 MAPK and bcl-2 in U251 glioma cells
AIM: To investigate the killing effects of Ad5F35-hIL-24 on U251 cells and its effect on p38 mitogen-activated protein kinase ( MAPK) pathway and bcl-2 protein expression. METHODS: Methyl thiazolyl tetrazolium (MTT) and double fluorescence staining were used to analyze the inhibition rate of U251 cells. Western Blot was applied to detect the protein expression of p38 MAPK, p-p38 MAPK and bcl-2. RESULTS: Ad5F3S-hIL-24 inhibited U251 cells in a dose-dependent manner. Ad5F35-hIL-24 increased p-p38 MAPK protein expression and decreased bcl-2 protein expression compared with those in control groups. CONCLUSION: p38 MAPK and bcl-2 play some important roles in IL-24 gene induced U251 cell apoptosis. These results are of some value for clinical treatment of glioma.
