维药西帕依溃结安对大鼠溃疡性结肠炎模型COX-2表达的影响

目的:研究维药西帕依溃结安(KIA)对溃疡性结肠炎模型大鼠结肠组织中环氧化酶2(COX-2)表达的影响.方法:采用2,4-二硝基氧苯与乙酸复合法制备大鼠溃疡性结肠炎模型,分为5组进行实验.应用RT-PCR和Western Blot 方法分别检测各组COX-2的mRNA和蛋白质表达水平.结果:KJA大剂量干预组COX-2的蛋白质表达水平与生理盐水阴性对照组相比均明显下调,差异有统计学意义(P<0.05);KIA中剂量干预组和5-氨基水杨酸治疗组的COX-2的mRNA表达水平与生理盐水阴性对照组相比明显下调,差异有统计学意义(P<0.05).结论:KJA对溃疡性结肠炎的治疗作用可能与下调COX-2的表达有关,而其调节COX-2的表达可能发生在转录水平上.
Effect of Xipayi Kuijie'an on COX-2 gene expression in rat ulcerative colitis
AIM: To study the effect of the Uygur medicine Kuijie'an (KJA) on the expression of cyclooxygenase-2 (COX-2) in intestinal tissues of rat with ulcerative colitis (UC). METHODS: UC Wistar rate induced by 2,4-dinitrochlorobenzene(DNCB) and acetic acid were divided into 5 groups. The methods of semi-quan-titation RT-PCH and Western Blot were used to detect the different expression of mRNAs and proteins in each group. RESULTS: In comparison with the normal saline treated negative control group (NS) , the expression levels of COX-2 protein were significantly down-regulated in the large-dosage group (P<0.05) and the expression levels of COX-2 mRNA were significantly down-regulated in the middle-dosage group and the amino salicylic acid positive control group (5-ASA) (P<0. 05). CONCLUSION: The down-regulation of COX-2 at the transcription level may contribute to the possible therapeutic action of KJA.
