Significance of Nanog gene expression in osteosarcoma.like tumor stem cells
AIM: To study the expression of Nanog gene in osteosarcoma-like tumor stem cells and its significance. METHODS: Tumor sphere cells were isolated from osteosarcoma cell line U2-OS and MG-63 with serum-free culturing techniques, Stro + cells were isolated using magnetic activated cell sorting system. RT-PCR was used to delect the expression of Nanog gene in Stro-1 + cells, Stro-1- cells and tumor sphere cells. RESULTS: The levels of Nanog mRNA expression in stro-1 + cells and tumor sphere cells were 1.69 ±0. 14 and 1.56 ±0.18 respectively, higher than that in the Stro-1- cells(P <0. 05). Expression of Nanog mRNA in sphere cells decreased with the prolongation of culture (P < 0.05). CONCLUSIONS: Nanog gene is highly expressed in osteosarcoma-like tumor stem cells, which might be helpful to sustain the self-renewal and the undif-ferentiation of osteosarcoma-like tumor stem cells.
