目的:建立广泛表达红色荧光蛋白(RFP)的转基因小鼠,便于组织和细胞移植研究.方法:把DsRed基因片段插入到pCAGGS载体的强启动子下游构建转基因载体pCAG-DsRed.经细胞转染验证,受精卵原核注射,建立红色荧光蛋白转基因C57BL/6小鼠.PCR鉴定转基因小鼠的基因型;红色荧光蛋白在组织中的表达情况,则通过荧光显微镜检测小鼠各组织器官冰冻切片.结果:pCAG-DsRed载体可以在真核细胞中表达RFP,经基因型分析和表型观察,得到两只红色荧光蛋白的首建鼠.经荧光显微镜观察各组织冰冻切片发现红色荧光蛋白在多个组织器官中表达,其中在神经系统和肌肉组织中表达量较高.结论:成功建立了一种红色荧光蛋白转基因小鼠,红色荧光蛋白在转基因小鼠的神经系统和肌肉组织等中高表达.
Establishment of red fluorescence protein transgenic mouse model
AIM: To establish a ubiquitously expressed red fluorescent protein (RFP) transgenic mouse model for tissue and/or cell transplantation. METHODS: The transgenic vector pCAGDsRed was constructed by inserting the DsRed fragment into the downstream of the strong promoter in the pCACGS vector and was validated by cell transfection. The BFP transgenic mouse model was established by pronuclear injection of fertilized eggs. The genotype of the transgenic mouse was confirmed by PCR. The expression patterns of the RFP in transgenic mice were detected by observations of frozen sections from all the tissues and organs under a fluorescent microscope. RESULTS: RFP could be expressed by the pCAG-DsRed vector-transfected eukaryotic cells. Two RFP transgenic founder mice on C57BL/6 background were generated and confirmed by PCB and tissue detections. It was found that the BFP was expressed in a number of tissues, especially in the nervous and muscle tissues. CONCLUSION: A line of RFP transgenic mouse model has been successfully established and the RFP highly expresses in the nervous and muscle tissues.
