Establishment of red fluorescence protein transgenic mouse model
AIM: To establish a ubiquitously expressed red fluorescent protein (RFP) transgenic mouse model for tissue and/or cell transplantation. METHODS: The transgenic vector pCAGDsRed was constructed by inserting the DsRed fragment into the downstream of the strong promoter in the pCACGS vector and was validated by cell transfection. The BFP transgenic mouse model was established by pronuclear injection of fertilized eggs. The genotype of the transgenic mouse was confirmed by PCR. The expression patterns of the RFP in transgenic mice were detected by observations of frozen sections from all the tissues and organs under a fluorescent microscope. RESULTS: RFP could be expressed by the pCAG-DsRed vector-transfected eukaryotic cells. Two RFP transgenic founder mice on C57BL/6 background were generated and confirmed by PCB and tissue detections. It was found that the BFP was expressed in a number of tissues, especially in the nervous and muscle tissues. CONCLUSION: A line of RFP transgenic mouse model has been successfully established and the RFP highly expresses in the nervous and muscle tissues.
