Experiment study of protective effects of combined Verapamil and Glutathione on apoptosis of allograft rabbit lung
AIM: To investigate protective effects of combined administration of Verapamil and Glutathione on apoptosis of allograft rabbit lung. METHODS: Twenty-four pairs of rabbits were randomly divided into 4 groups with 6 pairs in each: Croup Ⅰ as control in which lungs were flushed and preserved with modified LPD solution. Group Ⅱ, using modified LPD solution plus verapamil 2 mg/kg, Group Ⅲ, using modified LPD solution plus glutathione 3 mmol/L, and Group Ⅳ, using modified LPD solution plus verapamil 2mg/kg and glutathione 3 mmol/L. All lungs were reperfused for 2 h after 4 h cold preservation. Lung tissue MDA and SOD, intracellular Ca~(2+) and apoptosis were measured. RESULTS: Lung tissue MDA, intracellular Ca~(2+), mean gray value and Mascclina unit of apoptosis in group Ⅳ were lower than those in other three groups, lung tissue SOD, the mean gray value and Masccline unit of apoptosis in Bcl-2 in group Ⅳ were higher than those in other three groups. CONCLUSION: Preservation of allograft rabbit lung with combination of verapamil and glutathione is better than single administration for combined administration alleviates the apoptosis in lung transplantation.
