Effect of GPS on JAK_2-STAT_5, NF-κB signal transduction pathways in K562 cells
AIM: To investigate the effect of Ginseng polysaccharide (GPS) on JAK_2-STAT_5 and NF-κB signal transduction pathways in K562 cells. METHODS: We observed the expression of JAK_2, NF-κB and STAT_5 in GPS-induced K562 cells by techniques of immunocytochemistry and Western blot, and examined the effects of GPS on the phosphorylation of JAK_2 in K562 cells using immunoprecipitation. The expressious of c-myc and p53 in K562 cells induced by GPS were measured by BT-PCR. RESULTS : The expression of JAK_2 protein increased with the increasing concentration of GPS and the treatment time[ ( 12.33 ±2.05) vs (88.18±13.57)]. The expression of NF-κB p65 and STAT_5 protein markedly decreased,[ (79.48±9.56) vs (6.98 ±1.45)] and[ (71.56±16.28) vs (10.23±2.16)]. GPS stimulated the tyrosine phosphorylation of JAK_2. After induction of K562 cells by GPS, the expression of c-myc mRNA decreased [ (196±12)vs (136±11)], while no marked changes were observed in the expression of p53 mRNA[ (202 ± 16) vs ( 181±16) ]. CONCLUSION: GPS regulates the signal transduction pathways of JAK_2-STAT_5 and NF-κB and prohibits the expression of c-mye oncogene.
