Expression of angiotensin-converting enzyme 2 in various tissues of rats with metabolic syndrome
AIM: To determine the expressions of angiotensin-converting enzyme 2 (ACE2) in the heart,blood vessel endothe-lium,lungs,livers,pancreas,kidneys,testis and brain in the rats with metabolic syndrome. METHODS: Twenty adult male Wistar rats were divided into control group (normal diet + ordinary tap water n = 10) and fructose-induced MS group (normal diet + 30% -fructose solution n = 10). After 6 weeks,the level of trigly-eride (TG),total cholesterol (TC),high-density lipoprotein cholesterol ( HDL-C ),low-density lipoprotein cholesterol ( LDL-C ),insulin sensitivity index (ISI),visceral fat mass over body weight ratio and Lee's index were assessed,and oral glucose tolerance test (OGTT) was evaluated. The levels of mRNA expression of ACE2 were determined by real-time PCR and the distribution and expression of ACE2 in these tissues were determined by immunohistochemistry. RESULTS: The levels of TG [ ( 1. 18 ± 0.19) vs (1.74±0.12)mmol/L],TC [(1.44 ±0.13) vs (2.04 ±0.18)mmol/L],LDL-C [(0.39 ±0.09) vs (0.89 ±0.12) mmol/L],Lee's index[ (297.48 ±9. 82) vs (323.50 ± 12.95) ],visceral fat mass over body weight ratio [(0.0373 ±0.0019) vs (0.0442 ±0.0014)] and BP [(109. 34 ±5. 02) vs (144.47 ± 3.26)mmHg] markedly increased in MS group(P<0.05). Statistically significant reductions in HDL-C [(0.75 ±0.12) vs (0.43 ±0.10) mmol/L] and ISI [ ( - 4. 48 ± 0. 07) vs ( - 5. 26 ± 0.16) ] was observed in MS group compared with those in control group. OGTT abnormity in MS group was obvious [ (8.26 ±0.56) vs (9.22±0.59)mmol/L] (P<0.05). ACE2 mRNA in the heart [(0.97±0. 13) vs (0.55 ±0. 11)],blood vessel endothelium [(16.71 ±0.46) vs (8.53 ±0.28)],lungs[(31.11 ±0.24) vs (12.45 ±0.11)],kidneys[ (37.48 ±0.27) vs (15.64 ±0.15) ],testis [(4. 85 ±0.05) vs (1.78 ±0.15)] and brain [(24.23 ± 0.25) vs (4. 82 ± 0.15)] significantly decreased in MS rats compared with those of control group (P < 0. 05). ACE2 mRNA was not observed in the livers and pancreas. CONCLUSION: ACE2 mRNA in the heart,blood vessel endothelium,lungs,kidneys,testis and brain significantly decrease in the rats with metabolic syndrome.
